چکیده

Rab GTPases and their effectors regulate membrane traffic by determining, along with cognate SNAREs, the specificity of transport vesicle docking and fusion steps. Recent studies have also implicated Rabs in the movement of these transport vesicles from their site of formation to their site of fusion, and several Rabs have been linked to specific microtubuleor actin-based motor proteins. Analyses of Rab and motor protein mutants, coupled with advanced imaging techniques, have led to the suggestion that certain Rabs function as essential components of the vesicle receptor for specific motor proteins.

Rab Proteins and the Secretory Pathway: The Case of Rab18 in Neuroendocrine Cells

The secretory pathway is a process characteristic of cells specialized in secretion such as endocrine cells and neurons. It consists of different stages that are dependent on specific transport of proteins in vesicular-tubular carriers. Biochemical analyses have unveiled a number of protein families that confer identity to carrier vesicles and specificity to their transport. Among them is the f...

Regulation of Receptor Trafficking by Rab GTPases

Human cells contain more than 60 Ras-like, Rab GTPases that are localized to the surfaces of distinct membrane-bound compartments. Rabs are master regulators of membrane traffic events: they can catalyze the formation of membrane microdomains, the collection of cargo into transport vesicles, the motility of vesicles along cytoskeletal tracks, the docking of vesicles with their targets, and the ...

Rab11 in Disease Progression

Membrane/ protein trafficking in the secretory/ biosynthetic and endocytic pathways is mediated by vesicles. Vesicle trafficking in eukaryotes is regulated by a class of small monomeric GTPases the Rab protein family. Rab proteins represent the largest branch of the Ras superfamily GTPases, and have been concerned in a variety of intracellular vesicle trafficking and different intracellular sig...

Direct interaction between a myosin V motor and the Rab GTPases Ypt31/32 is required for polarized secretion.

Rab GTPases recruit myosin motors to endocytic compartments, which in turn are required for their motility. However, no Ypt/Rab GTPase has been shown to regulate the motility of exocytic compartments. In yeast, the Ypt31/32 functional pair is required for the formation of trans-Golgi vesicles. The myosin V motor Myo2 attaches to these vesicles through its globular-tail domain (GTD) and mediates...

Rab6-bound GTP (stick model) and magnesium (sphere) are shown. (Bottom) View of the Rab6-GCC185 binding interface. Residues in GCC185 that are critical for Rab6 interaction and Golgi localization are labeled. An anomalous differenc...
